Second Calvary Baptist Church was founded in 1889 by about three hundred Christians who initially gathered at the home of Mr. and Mrs. George Hull before moving to various locations. The Bull Street church was dedicated in 1891. Over the years, several ministers led the congregation, including Reverends D. Cooper, A. P. Dunbar, R.F. Lee, and others.

The current Bull Street building project began in 1956 and was completed in 1964, with the mortgage paid off by 1970. Reverend Dr. Latta R. Thomas served from 1975 to 1995, recognized for his scholarship and leadership. Afterward, Reverend Preston H. Winkler served as interim, followed by Reverend John N. Osborne, Jr., who served from 1998 to 2006 and oversaw significant organizational changes and initiated relocation to Mason Road.

Groundbreaking for the new location occurred in 2005, with final services at Bull Street in April 2006 and dedication of the new church in October 2006. Reverend Isaiah Harvey became Interim Pastor in 2007, encouraging spiritual recommitment. In late 2008, Reverend Carl R. Wells was elected pastor, leading membership growth and youth program development, and serving until May 2022. Reverend Alex Nelson graciously served as Interim Pastor continuing a legacy of biblical principles until March of 2024.

Reverend Darrel S. President, Sr. led from March to December 2024, focusing on digital presence and branding. In May 2025, Dr. Bobby Cunningham was appointed Interim Pastor as the search for a permanent shepherd continues.
